Cytogenetic and FISH Analyses of the Abortus Materials: A Useful Approach Towards Genetic Counseling for the Couples with Recurrent Spontaneous Abortions
نویسندگان
چکیده
Objective: The cytogenetic analyses of the abortus material provide important information about the recurrence risks and possible management for the couples with recurrent spontanous abortions. The methods of cytogenetic analyses depend on primarily cell culture that has relatively higher (1040%) failure. Since the most common chromosomal abnormalities in abortus materials are numerical chromosomal abnormalities; we evaluated the compatibility of the karyotypes and interphase-Fluoresan in situ Hybridization (FISH) results and the predictive value of interphase FISH technique in cases with tissue culture failure. Materials & Methods: Interphase-FISH slides were prepared using touch preparation protocol from fresh tissue samples to eliminate the collagenase procedure. FISH procedure was carried out using specific probes for centromeric 13, 16, 18 and 21. At least 100 interphase cells for each slides were detected. Results and Conclusion: Chromosomal abnormality incidence of our study (45.5%) seems to be compatible with literature. It is founded some numerical aberrations (chromosomal abnormalities) as 18.18%autosomal trisomies, 13.64% triploidies, 4.50% monosomies and 9% mosaicism in cases. Two out of twenty eight abortus materials with tissue culture failure were revealed trisomies by FISH. However, the tissue culture failure incidence of this study (56%) is higher than the reported incidences (10-40%). This shortcoming could be diminished by the improvement of the tissue procurement and transportation conditions for culture. With this approach, supportive data will be accumulated and utilized for future epidemiological studies by concurrent FISH preparations and culture set up. ©2004, Fırat Üniversitesi, Tıp Fakültesi

Chromosomal Analysis of Couples with Repeated Spontaneous Abortions in Northeastern Iran
Background Cytogenetic study of reproductive wastage is an important aspect in determining the genetic background of early embryogenesis. Approximately 15 to 20% of all pregnancies in humans are terminated as recurrent spontaneous abortions (RSAs). The aim of this study was to detect chromosome abnormalities in couples with RSAs and to compare our results with those reported previously. Materia...

The most significant complication of pregnancy is recurrent miscarriage. Numerous factors have been described as associations with recurrent wastage such as: uterine abnormalities, immunological factors, endocrinologic imbalance and chromosomal defects. Cytogenetic evaluation of couples with recurrent pregnancy losses is performed on the basis of G-banding technique only after other possible et...

Chromosomal analysis is an important etiological investigation in couples with repeated spontaneous abortions as it helps in genetic counseling and deciding about further reproductive options. Abortions ,specially first trimester abortions is a very common complication and a matter of concern for couples planning pregnancy. Balanced chromosomal rearrangements in either parent is an important ca...
متن کاملSubtelomeric Rearrangements in Patients with Recurrent Miscarriage
Objective The Subtelomeric rearrangements are increasingly being investigated in cases of idiopathic intellectual disabilities (ID) and congenital abnormalities (CA) but have also been suspected to be responsible for unexplained recurrent miscarriage (RM). We have noticed a higher risk of subtelomeric translocations in association with CA and ID. Such rearrangements can go unnoticed through con...

Abstract Background A vast variety of factors may cause recurrent pregnancy loss. Blood group incompatibility of parents could cause abortion. The examination of couples or twins, blood groups showed that the blood group incompatibility can affect adversely the outcome of pregnancy. Couples with blood group incompatibility are more involved in spontaneous miscarriage. Antigenes in two differe...
